Byla ur\u010dena p\u0159esn\u00e1 m\u00edsta odb\u011bru vzork\u016f a kontroln\u00ed m\u00edsta. Pr\u016fzkumy byly prov\u00e1d\u011bny ve dvou agrolesnick\u00fdch syst\u00e9mech: ve sm\u00ed\u0161en\u00e9m ovocn\u00e1\u0159sk\u00e9m syst\u00e9mu a ve sm\u00ed\u0161en\u00e9m o\u0159echov\u00e9m syst\u00e9mu. Kontroln\u00edm m\u00edstem pro syst\u00e9m s o\u0159echovou sm\u011bs\u00ed bylo p\u0159ilehl\u00e9 pole s krmnou plodinou, zat\u00edmco syst\u00e9m s ovocnou sm\u011bs\u00ed p\u0159edstavoval p\u0159ilehl\u00e9 pole bez v\u00fdsadby. B\u011bhem diskus\u00ed na m\u00edst\u011b bylo rozhodnuto, \u017ee protokol o odb\u011bru vzork\u016f p\u016fvodn\u011b vypracovan\u00fd v r\u00e1mci \u00fakolu 3.2 bude p\u0159izp\u016fsoben podm\u00ednk\u00e1m lokality. M\u00edsto gradientu se \u010dty\u0159mi vzd\u00e1lenostmi od strom\u016f byly zvoleny dv\u011b vzd\u00e1lenosti: jedna bezprost\u0159edn\u011b p\u0159il\u00e9haj\u00edc\u00ed ke strom\u016fm a druh\u00e1 uprost\u0159ed mezi nimi. Tato \u00faprava byla nutn\u00e1, proto\u017ee stromy byly vysazeny teprve rok nebo dva p\u0159ed zah\u00e1jen\u00edm monitoringu, tak\u017ee jejich vliv byl patrn\u00fd p\u0159edev\u0161\u00edm v mal\u00fdch vzd\u00e1lenostech. Nep\u0159\u00edm\u00e9 vzd\u00e1lenosti nem\u011bly b\u00fdt v tomto kontextu v\u00fdznamn\u00e9.<\/p>\n\n\n\n<p>V obou syst\u00e9mech a jejich p\u0159\u00edslu\u0161n\u00fdch kontrol\u00e1ch byly zalo\u017eeny zku\u0161ebn\u00ed pozemky o rozm\u011brech 30 x 10 metr\u016f. D\u00e9lka 30 metr\u016f byla zvolena proto, \u017ee p\u0159i odb\u011bru n\u011bkter\u00fdch vzork\u016f biodiverzity bylo nutn\u00e9, aby pozemky byly od sebe vzd\u00e1leny alespo\u0148 10 metr\u016f. Za p\u0159edpokladu, \u017ee by pro ka\u017edou variantu byly po\u017eadov\u00e1ny \u010dty\u0159i opakov\u00e1n\u00ed, byly by tyto um\u00edst\u011bny pod\u00e9l jednoho transektu ve vzd\u00e1lenostech 0 m, 10 m, 20 m a 30 m. \u0160\u00ed\u0159ka 10 metr\u016f byla zvolena kv\u016fli 20metrov\u00fdm rozestup\u016fm mezi \u0159adami strom\u016f. Odb\u011br vzork\u016f byl napl\u00e1nov\u00e1n tak, aby transekt vedl pod\u00e9l strom\u016f a dos\u00e1hl st\u0159edu orn\u00e9ho pole (viz obr. 1 a\u017e 3). Tento pr\u016fzkum byl prov\u00e1d\u011bn na t\u0159ech desetimetrov\u00fdch transektech na ka\u017ed\u00e9 studijn\u00ed plo\u0161e (obr. 5). Byla pou\u017eita specializovan\u00e1 entomologick\u00e1 s\u00ed\u0165 s metrovou n\u00e1sadou a otvorem ve tvaru srdce o rozm\u011brech 30 x 40 cm. Tato konstrukce usnadnila sb\u011br hmyzu jak z vysok\u00e9 vegetace, tak z povrchu bl\u00edzk\u00e9ho zemi. Sb\u011brn\u00fd vak s\u00edt\u011b je vyroben z jemn\u00e9 s\u00ed\u0165oviny vyztu\u017een\u00e9 pouzdrem z pevn\u00e9ho materi\u00e1lu (obr. 6). Hmyz nasb\u00edran\u00fd b\u011bhem pr\u016fzkumu byl p\u0159enesen do 100 ml n\u00e1dob, eutanazov\u00e1n a konzervov\u00e1n pomoc\u00ed etanolu 75% (obr. 7). Byly pou\u017eity dva typy past\u00ed: Barberovy zemn\u00ed pasti a \u017elut\u00e9 pasti na opylova\u010de. Tyto pasti byly rozm\u00edst\u011bny ka\u017ed\u00fdch 10 metr\u016f pod\u00e9l ur\u010den\u00fdch transekt\u016f, p\u0159i\u010dem\u017e na ka\u017ed\u00e9 variant\u011b byly t\u0159i pasti (obr. 8). Barberovy zemn\u00ed pasti o pr\u016fm\u011bru 10 cm byly navr\u017eeny tak, aby \u00fa\u010dinn\u011b zachytily pozemn\u00ed hmyz. \u017dlut\u00e9 pasti na opylova\u010de o pr\u016fm\u011bru 30 cm byly um\u00edst\u011bny p\u0159\u00edmo na zem z d\u016fvodu n\u00edzk\u00e9 v\u00fd\u0161ky vegetace. Oba typy past\u00ed byly napln\u011bny do dvou t\u0159etin roztokem \u010dist\u00e9 vody a sapon\u00e1tu bez z\u00e1pachu (obr. 9). Toto slo\u017een\u00ed roztoku zaji\u0161\u0165ovalo rychl\u00e9 utopen\u00ed bezobratl\u00fdch a z\u00e1rove\u0148 zabra\u0148ovalo p\u0159elit\u00ed p\u0159i siln\u00e9m de\u0161ti, p\u0159i\u010dem\u017e vzhledem ke kr\u00e1tk\u00e9 dob\u011b sb\u011bru nebylo t\u0159eba pasti p\u0159ikr\u00fdvat. V dob\u011b sledov\u00e1n\u00ed nebyl z\u00edsk\u00e1n v\u00fdnos z p\u0159\u00edtomn\u00fdch strom\u016f, ani nebyla oseta obiln\u00e1 pole. D\u00e1le byla provedena fotodokumentace jednotliv\u00fdch druh\u016f rostlin v ka\u017ed\u00e9m syst\u00e9mu a jejich kontrola (obr. 13). Tyto fotografie byly nahr\u00e1ny do syst\u00e9mu iNaturalist pro ov\u011b\u0159en\u00ed identifikace druh\u016f, kter\u00fd vyu\u017e\u00edv\u00e1 algoritmy a konzultace s odborn\u00edky. Ka\u017ed\u00fd vzorek se skl\u00e1dal z nejm\u00e9n\u011b p\u011bti jader odebran\u00fdch vzorkova\u010dem Egner (obr. 23). Vzorky byly odeb\u00edr\u00e1ny do hloubky 30 cm. Po odb\u011bru byly vzorky p\u016fdy ulo\u017eeny do lednice a po n\u00e1vratu z ter\u00e9nn\u00ed exkurze hluboce zmrazeny.
